ABSTRACT Objective The larvae death caused by the infection of Micropterus salmoides rhabdovirus (MSRV) has seriously affected development Largemouth Bass nigricans industry. Therefore, it is necessary to establish a fast, convenient, and sensitive detection method detect MSRV-positive fish quickly control virus transmission. Methods In this study, primers probes were designed based on G gene MSRV. Through primer screening, specificity, sensitivity, repeatability tests, reverse transcription multienzyme isothermal rapid amplification (RT-MIRA) combined with lateral flow dipstick (LFD) was established. Additionally, we applied published transcription-polymerase chain reaction simultaneously clinical samples. Results temperature 42°C, total duration entire process 35 min. specificity test revealed that MSRV could be specifically detected had no cross-reaction other common susceptible pathogens fish. sensitivity indicated limit for recombinant plasmid standard 102 copies/μL. repetitive showed good reproducibility. There significant difference between results RT-MIRA-LFD RT-PCR. Conclusions established in study detecting simple, highly sensitive, specific, without need precise or expensive equipment. suitable resource-limited laboratories on-site detection.
