The degree of yellowing in tobacco leaves is an important indicator for determining the maturity and harvesting time leaves. Decreasing chlorophyll levels helps speed up ripening process easier mechanical harvesting. Identifying utilizing genes that regulate are crucial developing varieties suitable mechanized understanding molecular processes control leaf color changes. In this study, phenotypes yellow-leaf K326 were analysed, it was observed variety exhibited a distinct yellow phenotype with significant reduction content. Subsequently, using combination BSA-seq, transcriptomic sequencing (RNA-seq), proteomic approaches, we identified candidate gene Nitab4.5_0008674g0010 encodes dihydroneopterin aldolase as factor associated yellowing. Finally, by measuring folate content Huangye K326, to be significantly lower than thus indicating synthesis plays role phenotypic changes This study first use BSA-seq combined RNA-seq identify results provide theoretical basis analysis mechanism mutations.
