Toll-like receptors (TLRs) have been implicated in the regulation of various metabolism pathways, addition to their function innate immunity. Here, we investigate metabolic TLR2 a larval zebrafish system. We studied larvae from tlr2 mutant and wild type sibling controls an unchallenged normal developmental condition using transcriptomic metabolomic analyses methods. RNAseq was used evaluate differences between wild-type control found signature set 149 genes be significantly altered gene expression. The expression level several confirmed by qPCR analyses. Gene enrichment analysis (GSEA) revealed differential two genotypes related valine, leucine, isoleucine degradation glycolysis gluconeogenesis. Using 1H nuclear magnetic resonance (NMR) metabolomics, that glucose metabolites with were present at higher levels mutant. Furthermore, is mutants fluorometric assay. Therefore, shown TLR2, its immunity, has controlling during vertebrate development. functions are associated transcriptional enzymes involved could explain different glucose, lactate, succinate, malate

Rapid ascent to high-altitude areas above 2500 m often leads acute high altitude illness (AHAI), posing significant health risks. Current models for AHAI research are limited in their ability accurately simulate the environment drug screening. Addressing this gap, a novel static self-assembled water vacuum transparent chamber was developed induce zebrafish. This study identified 6000 2 h as optimal condition induction Under these conditions, notable behavioral changes including slow movement, abnormal exploration behavior and Novel tank test. Furthermore, model demonstrated oxidative stress-related markers included increased levels of malondialdehyde, decreased glutathione, activities superoxide dismutase catalase, inflammatory IL-6, IL-1β TNF-α, cell infiltration mild edema gill tissue, mirroring clinical pathophysiology observed patients. innovative zebrafish not only offers more accurate representation but also provides high-throughput platform discovery pathogenesis research.

Chloramine T, a sodium p-toluene sulfonchloramide, is known to possess wide spectrum of biocidal activity and employed as disinfectant in fish farms treat bacterial infections. Although T may effectively combat pathogens, the sublethal lethal effects changes acetylcholinesterase (AChE) remain poorly elucidated using Danio rerio (zebrafish) embryos. Zebrafish considered model organism for toxicant screening research exhibits mammalian-like physiological responses when exposed environmental pollutants. The aim this study was (1) determine LC50 after 96 hr exposure, (2) verify on developmental morphology, (3) evaluate AChE zebrafish exposure performed 16, 32, 64, 128, or 256 mg/L concentrations. mortality values were 143.05 ± 3.11 130.97 7.4 at 24 hr, respectively. Data demonstrated delayed hatching, reduced heartbeats, cardiac edema, equilibrium disruption hatched larvae throughout embryonic development. In addition, inhibited 64 128 treatment, corroborating sub-lethality results observed embryo development demonstrating an larvae.

Data variability complicates reproducibility and the interpretation of experimental results. Different animal models have been employed to decrease enhance power. However, variation frequently persists among within strains/lines. In zebrafish (Danio rerio), inbred lines (e.g., NHGRI-1) derived from wild-type produced greatly genetic variation, with goal providing better understanding backgrounds that may influence outcome studies employing such lines. We hypothesized variations in morphological phenotypes shaped by environmental stressors early development are correlated intrinsic degree compared (yolk–chorion ratio, body mass, embryo total length, condition factor, specific growth rate) AB NHGRI-1 as a function their responses altered temperature oxygen availability during first 7 days post-fertilization. Overall, both showed similar developmental trajectories for yolk–chorion length. Additionally, factor rate each line, regardless hypoxia. Importantly, coefficient variable was significantly lower than larvae 151 187 assessed endpoints. Thus, low-heterozygosity line can be useful decreasing inter-individual stressors, thereby aiding results enhancing reproducibility.

Two aquatic ambient factors, viz. Dissolved Oxygen (DO) and pH were selected for the study on effect of oxidative stress in liver zebrafish. The zebrafish exposed to four DO saturation levels (20-30%, 40-50%, 60-70% 80% above) five (4.5-5.5, 5.5-6.5, 6.5-7.5, 7.5-8.5 8.5-9.5) different periods (DO, 4h-16h at 4hr interval; pH, 1h-4h 1 hr interval). antioxidants studied catalase, glutathione superoxide dismutase (SOD). malondialdehyde (MDA) was assessed as a bioindicator stress. Results showed that fish suffered from all (except above, control) control). However, highest generation MDA observed 20-30% level 4.5-5.5 level. Under level, 12 h 2 treatments. Accordingly, antioxidant enzymes SOD also low activity treatment h, respectively. It evident alkaline (8.5-9.5), although altered activity, but not high 4.5-5.5, compared control.
