All species from order Actiniaria (sea anemones) are venomous, even though most of no threat to humans. Currently, we know very little about the toxin gene complement highly venomous members this order. To address gap in knowledge, sequenced transcriptome and medically significant Hell's Fire sea anemone, Actinodendron plumosum, as well five distantly related species, Cryptodendrum adhaesivum, Epiactis australiensis, Heteractis aurora, Isactinia olivacea Stichodactyla mertensii. We used bioinformatic approaches identify their complements performed a comparative evolutionary analysis seven understudied families. Of 16 families identified, 12-40 candidate toxins were found six new anemone transcriptomes, with only 12 candidates eight identified A. plumosum. Across 26 neurotoxin showed evidence taxonomic restriction, whereas phospholipase A2 family was ubiquitously distributed. Additionally, two alternative forms for family, 10- 14-cysteine framework, which warrant further structural functional characterisation. Overall, have comprehensive list wide diversity that provides basis future research structurally functionally characterise novel potential use therapeutics or agricultural applications.
