Anthocyanins, flavonoids with strong antioxidant activity, give rise to a fruit peel color and provide health benefits. Studies have revealed that melatonin promotes anthocyanin accumulation. However, the function of endogenous hormones in this process remains elusive. Here, we investigated effect ethylene signaling on accumulation melatonin-treated pear fruit. Melatonin promoted while repressing production; its biosynthesis background ethephon pretreatment was eliminated. Transcriptome analysis PuERF27, melatonin-induced response factor (ERF), as candidate gene. Transient overexpression PuERF27 is essential for enhancing activating genes, whereas silencing repressed Yeast one-hybrid, electrophoretic mobility shift, GUS assays indicated could bind directly positively regulate PuMYB10 PuGSTF12 promoters. Further studies together PuMYB10, synergistically transactivates promoter. These results indicate may promote expression indirectly activate structural genes through thereby leading transport anthocyanins.
