Endometriosis can cause of infertility, and evaluation methods for predicting clinical pregnancy outcomes are desired. Extracellular vesicles (EVs) exist in blood it contains small non-coding RNAs (ncRNAs) that may reflect disease severity. In this study, we investigated ncRNAs serum EVs to identify specific biomarkers pregnancy. Serum samples were collected from 48 patients who underwent assisted reproductive technology (ART). successfully isolated characterized using nanoparticle tracking assays, electron microscopy, western blotting EV's markers. We performed RNA sequencing analyzed microRNA (miRNA) profiles the infertility with without endometriosis detect pregnancy-predicting biomarkers. Candidate miRNAs selected by comparing became pregnant (n = 13) those did not 21). A total 241 detected; however, no trends separated two groups. Next, divided into 4) non-pregnant 10) cases. Among 224 candidate miRNAs, miRNA women receiver-operating characteristics (ROC) curve analysis (area under [AUC] > 0.8). endometriosis, be useful possible before treatment. Finally, used tissue demonstrate produced lesions. Although predictors found could provide novel noninvasive predict have potential applicability ART. Endometriosis is a chronic, estrogen-dependent disorder associated with the presence of endometrial cells mainly in pelvic cavity, causing systemic immune inflammation, infertility, epigenetic dysregulation differential DNA methylation, coelomic metaplasia, and pain. It affects approximately 10-12% women. Despite decades research, full pathophysiology, diagnostic roadmap, clinical management strategies for endometriosis are not yet fully elucidated. Cell-free (Cf-DNA) peripheral blood diseased healthy individuals was discovered 1950s. Quantifying Cf-DNA specific methylation group genes have been proposed as potential non-invasive biomarkers somatic constitutional genetics various other pathological disorders. In this study, we investigated levels 78 young women, 38 whom had confirmed via laparoscopy 40 were healthy. We found significant difference between two groups when quantified, 3.9 times more serum women endometriosis. also identified nine target potentially involved pathogenesis endometriosis, different profile groups. Our data suggest that combination cell-free quantification assessment signature can be predictive test

Emerging evidence suggests that aberrantly expressed microRNAs (miRNAs) participate in endometriosis pathogenesis. miR-1229-5p participates the pathogenesis of several disease, but its precise role and mechanism is unclear. Endometrial tissues were obtained from patients with healthy controls. RT-qPCR western blotting employed to detect expression levels genes proteins, respectively. Transcriptome sequencing luciferase reporter assay utilized identify target miR-1229-5p. CCK-8, transwell assay, wound healing flow cytometry performed evaluate functional roles Finally, clinical significance was furtherly analyzed. MiR-1229-5p upregulated ectopic endometrium ovarian (n = 60) compared normal endometria controls 40), also served as an indicator for severity. STMN1 identified by experiments, significantly downregulated endometrium. Functionally, overexpression promoted migration, invasion, inhibited apoptosis ESCs Ishikawa cells. Meanwhile, upregulation facilitated protein Bcl-2, MMP2, MMP9, N-cadherin, ZEB1, repressed Bax E-cadherin. Whereas downregulation exerted opposite effects. Importantly, could partially reverse effects upregulation. Mechanistically, activates p38 mitogen-activated kinase (p38 MAPK) signaling via targeting STMN1. The newly miR-1229-5p-STMN1-p38 MAPK axis illustrates molecular progression offers a potential therapeutic treating endometriosis.
