Abstract Seminal fluid protein composition is complex and commonly assumed to be rapidly divergent due functional interactions with both sperm the female reproductive tract (FRT), of which evolve rapidly. In addition sperm, seminal may contain structures, such as mating plugs spermatophores. Here, we investigate evolutionary diversification a lesser‐known ejaculate structure: spermatostyle, has independently arisen in several families beetles true bugs. We characterized spermatostyle proteome, FRT morphology, six species whirligig (family Gyrinidae). Spermatostyles were enriched for proteolytic enzymes, assays confirmed they possess activity. Sperm‐leucylaminopeptidases (S‐LAPs) particularly abundant, their localization spermatostyles was by immunohistochemistry. Although there evidence conservation proteomes across species, phylogenetic regressions suggest covariation between morphology FRTs. postulate that S‐LAPs (and other proteases) have evolved novel structural role discuss adaptations delivering male‐derived materials females.
