Planctopirus limnophila belongs to the bacterial phylum Planctomycetes, a relatively understudied lineage with remarkable cell biology features. Here, we report genome-wide analysis of essential gene content in P. limnophila. We show that certain genes involved peptidoglycan synthesis or division, which are most other studied bacteria, not for growth under laboratory conditions this species. identify likely lipopolysaccharide biosynthesis, consistent view Planctomycetes as diderm and highlight unknown functions. Furthermore, explore potential stages evolution repertoire related phyla Verrucomicrobia Chlamydiae. Our results provide insights into divergent molecular cellular Planctomycetes.

Pyrophosphatases (PPases) are enzymes that catalyze the hydrolysis of pyrophosphate (PPi), a byproduct synthesis and degradation diverse biomolecules. The accumulation PPi in cell can result death. Although substrate is same, there variations catalysis features these enzymes. Two enzyme forms have been identified bacteria: cytoplasmic or soluble pyrophosphatases membrane-bound pyrophosphatases, which play major roles bioenergetics. In eukaryotic cells, predominant form PPases (c-PPases), while membrane (m-PPases) found only protists plants. study bacterial has slowed recent years. These central to metabolism physiology since phospholipid nucleic acid release important amounts must be removed allow biosynthesis continue. this review, two aims were pursued: first, provide insight into structural known date well characterized, examples with novel features. Second, scientific community should continue studying because they many biotechnological applications. Additionally, we evidence m-PPases present fungi; date, no characterized. Therefore, diversity PPase still fruitful field research. focused on H + /Na pumps Finally, some applications molecular biology biotechnology, especially This review valuable for professionals biochemistry protein structure–function relationships experts other fields, such as chemistry, nanotechnology, plant sciences.

ABSTRACT The peptidoglycan (PG, or murein) is a mesh-like structure, which made of glycan polymers connected by short peptides and surrounds the cell membrane nearly all bacterial species. In contrast, there no PG counterpart that would be universally found in Archaea but rather various are specific to some lineages. Methanopyrales Methanobacteriales two orders Euryarchaeota harbor pseudomurein (PM), structural analog PG. Owing differences between PM biosynthesis, have argued origin both not connected. However, recent studies revealed genomes PM-containing encode homologs genes involved even though neither their functions nor relationships within corresponding inter-domain phylogenies been investigated so far. this work, we devised pangenomic bioinformatic pipeline identify proteins for biosynthesis without prior genetic knowledge. taxonomic distribution evolutionary candidate were studied detail Bacteria through HMM sequence mining phylogenetic inference Mur domain-containing family, ATP-grasp superfamily, MraY-like family. The microorganisms responsible for driving the Anammox and Sammox processes may have ancient origins remain poorly understood. In this research, we enriched from paddy soils marine sediments under conditions to investigate shifts in their structure functions. community showed significant differences between enrichment cultures of conditions. Among samples soils, Proteobacteria, Nitrospirota, Verrucomicrobiota, Gemmatimonadata, Euryarchaeota accounted a larger proportion culture conditions, while Firmicutes, Bacteroidota, Chloroflexi, Planctomycetota, Acidobacteriota, Crenarchaeota, Nanoarchaeota sediments, Genes associated with assimilatory sulfate reduction, nitrate denitrification pathways were more abundant whereas genes related dissimilatory reduction nitrification prevalent These findings provide valuable insights into ecological microbial species primordial era aid searching extraterrestrial life.

Abstract Plasmids, despite their critical role in antibiotic resistance and modern biotechnology, are understood only a few bacterial groups terms of natural ecological dynamics. The phylum Planctomycetes, known for its unique molecular cellular biology, has largely unexplored plasmidome. This study offers thorough exploration the diversity plasmids within which could serve as foundation developing various genetic research tools this phylum. Planctomycetes encode broad range biological functions appear to have coevolved significantly with host chromosomes, sharing many homologues. Recent transfer events insertion sequences between cohabiting chromosomes were also observed. Interestingly, 64% plasmid genes distantly related either chromosomally encoded or homologues from other groups. planctomycetal plasmidome is composed 36% exclusive proteins.
