Abstracts
The
large-scale
loach
(
Paramisgurnus
dabryanus
;
Cypriniformes:
Cobitidae)
is
primarily
distributed
in
East
Asia.
It
an
important
economic
fish
species
characterized
by
fast
growth,
temperature-dependent
sex
determination
and
the
ability
to
breathe
air.
Currently,
molecular
mechanism
studies
related
some
aspects
such
as
determination,
toxicology,
feed
nutrition,
growth
genetic
evolution
have
been
conducted.
However,
lack
of
a
high-quality
reference
genome
has
hindered
further
research.
In
this
study,
we
performed
PacBio
HiFi
Hi-C
sequencing
on
female
(ZW)
specimen
assembled
first
chromosome-level
loach.
comprises
24
chromosomes,
with
total
length
1.04
Gb,
scaffold
N50
41.7
Mb,
BUSCO
completeness
95.8%,
including
28,311
protein-coding
genes.
These
findings
not
only
provide
new
insights
into
structure
but
also
establish
crucial
point
for
omics
serve
essential
genomic
resource
breeding
programs
species.

Abstract
Early
antiretroviral
therapy
(ART)
initiation
is
known
to
limit
the
establishment
of
HIV
reservoir,
with
studies
suggesting
benefits
such
as
a
reduced
number
infected
cells
and
smaller
latent
reservoir.
However,
long-term
impact
early
ART
on
dynamics
cell
pool
remains
unclear,
clinical
evidence
directly
comparing
proviral
integration
site
counts
between
late
limited.
In
this
study,
we
used
Linear
Target
Amplification-PCR
(LTA-PCR)
Next
Generation
Sequencing
compare
unique
(UIS)
clonal
individuals
who
initiated
during
acute
infection
stage
(Acute-ART
group)
those
in
AIDS
(AIDS-ART
group).
Our
analysis
revealed
distinct
distribution
patterns,
greater
UIS
heterogeneity
Acute-ART
group
more
homogeneity
AIDS-ART
group.
Monoclonal
accumulation,
predominantly
in-gene
regions,
was
influenced
by
timing
duration,
treatment
delaying
process.
Host
genes
integrated
provirus
monoclonal
types
were
enriched
cycle
lymphocyte
activation
pathways.
Tumor
suppressor
(TSGs)
frequently
group,
potential
risk
factors.
Overall,
introduced
sequencing
method
assess
size
human
peripheral
blood
identified
widespread
presence
after
treatment.
The
intervention
helps
slow
progress
expansion
cells,
reducing
formation
stable
persistent
reservoirs,
ultimately
posing
fewer
barriers
achieving
functional
cure.
