The
innovative
HaloTag
is
a
labeling
technology
that
plays
crucial
role
in
advanced
fluorescence
imaging.
However,
due
to
the
complexity
of
plant
materials,
application
field
plants
still
its
infancy.
To
expand
cells,
we
constructed
six
eukaryotic
gene
expression
vectors
with
different
localizations
carrying
labels,
then
transformed
Arabidopsis
thaliana
and
Nicotiana
benthamiana,
finally
stained
living
cells
using
ligands.
Live-cell
imaging
showed
did
not
affect
localization
pattern
biological
functions
target
protein,
by
improving
experimental
conditions,
optimized
could
be
more
effectively
applied
plants.
Remarkably,
first
displayed
exhibits
superior
performance
compared
conventional
fluorescent
proteins,
including
photostability,
monomer
properties,
multicolor
spatio-temporal
differentiated
labeling.
More
importantly,
combination
single-particle
tracking
reveals
specific
dynamics
membrane
proteins
changes
rearrangement
ER-PM
connectivity,
providing
powerful
tool
for
analyzing
protein
dynamics.
Taken
together,
this
promising
enriches
toolkit
studying
spatiotemporal
various
processes.
