ABSTRACT Anthocyanins (Anthos; flower and kyanos; blue) are natural coloring compounds from the flavonoids class that include cyanidin, peonidin, delphinidin, malvidin, pelargonidin, petunidin. Recently, role of anthocyanins in disease prevention, especially inflammation, diabetes, cancer, neuro‐disorders, hepato‐renal protective, immuno‐modulation properties has been highlighted. The current review covered literature on pharmacokinetics pharmacological effects anthocyanins, absorption, distribution, metabolism, excretion (ADME). discussion molecular mechanisms underlying their therapeutic is limelight article. GLUT1, GLUT3, SGLT1, SMCT1, SMCT2 main carriers involved transportation gastrointestinal tract. exert anticancer by reducing expression IL‐6, IL‐1β, TNF‐β, COX‐2, downregulation NF‐kB, EZH2, MDR1, Akt , modulation P13K/AKT AMPK/mTOR pathways. reduction α‐amylase α‐glucosidase improved FFAR1 activity results antidiabetic effects. regulation PGC‐1α/NRF2/TFAM, p‐PI3K/Akt/GSK3β, Nrf2/HO‐1 prevents neurodegeneration. impose protective via ameliorating NLRP3 inflammasome, inhibiting MDA, GSSG, iNOS, HO‐1, ICAM‐1, β2‐microglobulin, MPO activity, SOD, CAT, GSH activity. promote beneficial gut microbiota enhance SCFA production, thus pro‐inflammatory markers. immuno‐modulatory impact involves CRP, P‐selectin, C1q, C4. reduce LDL, VLDL, TGs, TC GBA upregulation ATP6 V0C, ZO‐1, ATG4D expression. WHO/FAO suggested 2.5 mg/kg/day grape‐skin extracts safe, China recommended 50 mg/day safe for consumption. In a nutshell, multifaceted health benefits make them promising candidates prevention interventions.
