Abstract Dibutyl phthalate (DBP), di-2-ethylhexyl (DEHP), and benzyl butyl (BBP) are used in personal medical care products. In the ovary, antral follicles essential for steroidogenesis ovulation. DBP, BBP, DEHP known to inhibit mouse follicle growth ovulation vitro, associate with decreased counts women. Given that vivo effects of a three-phthalate mixture on unknown, we evaluated human-relevant ovarian through proteome profiling analysis. Adult CD-1 female mice were fed corn oil (vehicle), or two dose levels based estimated exposures general (32 µg/kg/d; PHT 32) occupationally exposed (500 500) populations 10 d. Antral (>250 µm) isolated subjected via label-free tandem mass spectrometry. A total 5,417 proteins detected, which 194 differentially abundant between vehicle 32, 136 500. Bioinformatic analysis revealed significantly different responses doses. Protein abundance differences 32 exposure mapped cytoplasm, mitochondria, lipid metabolism; whereas those 500 nucleus, phosphorylation. When both doses altered common processes, associated predicted transcription factors different. These findings provide novel mechanistic insight into phthalate-associated, ovary-driven reproductive outcomes

Obesity and ovotoxicant exposures impair female reproductive health with greater ovotoxicity reported in obese relative to lean females. The mother developing fetus are vulnerable both during gestation. 7,12-dimethylbenz[a]anthracene (DMBA) is released carbon combustion including from cigarettes, coal, fossil fuels, forest fires. This study investigated the hypothesis that diet-induced obesity would increase sensitivity of ovaries DMBA-induced determined impacts DMBA exposure gestation on maternal ovary. Female C57BL/6 J mice were fed a control or High Sugar Fat (45% kcal fat; 20% sucrose) diet until ~30% weight gain was attained before mating unexposed males. From Day 7, exposed intraperitoneally either vehicle (corn oil) (1 mg/kg diluted corn for 7 d. Thus, there four groups: (LC); exposed; control; exposed. Gestational decreased (P < 0.05) ovarian increased liver weights LC dams, but no treatment impact > spleen progesterone. Also, exacerbated reduction number primordial, secondary follicles, corpora lutea. In mice, altered abundance 21 proteins; affected 134 proteins while alone 81 ovary impacted by metabolic status influences outcome.
