Abstract Anthocyanins are an essential class of flavonoids that represent a large group plant secondary metabolites. microRNAs (miRNAs) can target TFs related to anthocyanin synthesis and inhibit their expression, thereby affecting the expression level key downstream structural genes ultimately regulating accumulation anthocyanins in plants. Nevertheless, regulatory function miR156a synthesis, especially potatoes, has yet be investigated. In this study, its gene StSPL9 were screened analyzed from small RNA sequencing, degradome sequencing transient assay. The potato tuber was Overexpression (OE-miR156a) tubers promoted while concurrently reducing flavonoid compared with wild-type (WT) potatoes. miR156a-silenced (STTM-miR156a) achieved using short tandem mimics (STTM) contained significantly lower content increased WT OE-miR156a. Notably, overexpression yielded identical results. relative levels anthocyanin-related PAL , 4CL CHS CHI F3H DFR UFGT STTM-miR156a transgenic showed opposite trend observed OE-miR156a tubers, as demonstrated by quantitative real-time PCR (qRT-PCR). integrated analysis transcriptome metabolome revealed flavonols than wild type, whereas contents phenolic acids notably reduced. Further FG2 BEATH CYP75A up-regulated STTM-miR156a, HCT downregulated, which resulted reduced methyl 4-caffeoylquinate, acid metabolite. Therefore, it is indirectly modulates targeting degrading other SPL genes, thus influencing tuber. This study provides valuable insights into complex network governing biosynthesis Solanum species.
