Abstract Prenatal cannabis use is associated with adverse offspring neurodevelopmental outcomes, however the underlying mechanisms are relatively unknown. We sought to determine impact of chronic delta-9-tetrahydrocannabinol (THC) exposure on fetal neurodevelopment in a rhesus macaque model using advanced imaging combined molecular and tissue studies. Animals were divided into two groups, control (n = 5) THC-exposed 5), which received daily THC edible pre-conception throughout pregnancy. Fetal T2-weighted MRI was performed at gestational days 85 (G85), G110, G135 G155 assess volumetric brain development. At G155, animals underwent cesarean delivery collection cerebrospinal fluid (CSF) for microRNA (miRNA) studies histologic analysis. significant age by sex interactions growth, differences histology suggestive dysregulation. Two extracellular vesicle associated-miRNAs identified CSF; pathway analysis suggests that these miRNAs dysregulated axonal guidance netrin signaling. ABSTRACT Introduction Legalization in many jurisdictions has increased the prevalence of cannabis use, including during pregnancy and lactation. Accordingly, clinicians providing perinatal infant care are increasingly required to counsel about this topic, even if they do not feel comfortable or prepared for conversation. The aim research was explore how prenatal pregnant lactating women interact with consumption. Methods Using qualitative description, we conducted semi‐structured interviews 75 individuals Canada: 23 who provide lactation care, 52 made consumption decisions and/or Data were analyzed using inductive content analysis. Results Three phases clinical encounter influenced decision‐making consumption: initiation a discussion cannabis, sense‐making, outcome encounter. Patients described similar ideals counseling lactation: open, patient‐centered conversation grounded an informed model benefits, risks, alternatives cannabis. While these values as reflecting real interactions, patients reported that their experience, actual interactions did live up ideals. Cannabis is the most commonly used federally illicit substance during pregnancy in United States, with an estimated prevalence of 3% to 30%. The American College Obstetricians and Gynecologists Academy Pediatrics discourage cannabis use breastfeeding due safety concerns for fetus infant. Despite these recommendations, nearly half active users continue pregnancy. The aim of this study is to determine factors associated with cannabis discontinuation, assess the impact mental health and addiction interventions on discontinuation during pregnancy investigate neonatal discontinuation.
