Bacteria infecting the plant phloem represent a growing threat worldwide. While these organisms often resist in vitro culture, they multiply both sieve elements and hemipteran vectors. Such cross-kingdom parasitic lifestyle has emerged diverse taxa via distinct ecological routes. In genus Arsenophonus , pathogens 'Candidatus phytopathogenicus' (Ap) 'Ca. Phlomobacter fragariae' (Pf) have evolved from insect endosymbionts, but genetic mechanisms underlying this transition not been explored. To fill gap, we obtained genomes of strains host metagenomes. The resulting assemblies are highly similar size functional repertoire, rich viral sequences, closely resemble several facultative endosymbiotic sap-sucking hemipterans. However, phylogenomic analysis demonstrated origins, as Ap belongs to 'Triatominarum' clade whereas Pf represents distant species. We identified set orthologs encoded only by genus, including hydrolytic enzymes likely targeting substrates. particular, bacteria encode cell-wall degrading cysteine peptidases related xylellain, papain-like peptidase Xylella fastidiosa for which close homologs found proteobacteria vasculature. silico predictions expression analyses further support role during colonization shared orthologs. conclude that double emergence phytopathogenicity may mediated few horizontal gene transfer events, involving genes first acquired other phytopathogens.
