ABSTRACT In vertebrates, auditory information is transduced in the cochlea by mechanosensory hair cells (HC) through an eccentrically organised structure known as bundle. This consists of a true cilium, kinocilium, and modified microvilli, stereocilia. The bundle has distinct with stereocilia graded rows, longest abutting kinocilia. bundles all HC are aligned to tissue axis planar polarised. Important development physiology protein bridges consisting cadherin-23 protocadherin-15. These link tips stereocilia, where they play role mechanotransduction, between kinocilia involved development. Both protocadherin-15 mutations result defects polarity; however, mechanism which this defect arises unclear. Using novel mutant for Pcdh15-CD2 isoform, we show that while initial deflection kinocilium occurs, its peripheral migration register Gαi3 perturbed. genetically interacts Gpsm2, perturbing vestibular function. We find earliest expression at base kinocilia, morphogenesis occur before formation kinocilial links. By re-introducing functional Pcdh15-CD2, polarity can be restored, suggesting couples stereocilial independent
