A molecular atlas of human granulopoiesis
Abstract
We
developed
a
comprehensive
molecular
atlas
of
human
granulopoiesis,
encompassing
the
transcriptome
(mRNA),
proteome,
and
non-coding
RNA
(miRNA,
lncRNA)
seven
defined
stages
neutrophil
maturation.
Two
waves
mRNA
transcription
activity
were
observed.
Whereas
first
wave,
extending
from
myeloblasts
to
metamyelocytes,
showed
correlated
transcripts
protein
translation,
second
wave
was
characterized
by
uncoupling
translation.
Integrated
analysis
revealed
intricate
dynamics
translation
parallelization
in
granule
subset
proteins,
ribosomes,
mitochondria,
respectively.
identified
hsa-miR-106a-5p
family
hsa-miR-125a-5p
as
potential
repressors
myeloid
differentiation
myeloblasts,
hsa-miR-193a-5p
repressor
SWI/SNF
complex.
LncRNA
displayed
remarkable
stage
specificity.
NEAT1
MAILR
expressed
only
band
onwards,
ITGB2-AS1
exclusively
metamyelocytes.
Novel
lncRNAs
with
high
expression
found
terminal
S-stage
PMN
(ENSG00000288819,
ENSG00000203644).
A
factor
signatures,
including
on
target
genes,
previously
unrecognized
factors
well
yet
underappreciated
transcriptional
role
lactotransferrin
(LTF)
early
maturation
stages.
Characterization
single-cell-clusters
via
profiles
histologically
enabled
stage-specific
cell
labeling.
