Abstract Mitochondrial stress activates the integrated response (ISR) and triggers cell-cell communication through secretion of metabokine growth differentiation factor 15 (GDF15). However, gene network underlying ISR remains poorly defined, particularly across metabolically diverse cellular states tissues. Using RNAseq data from fibroblasts subjected to metabolic perturbations, we develop an GDF15 index quantifying arm activation in human cells. Validation 44 postmortem tissues illustrates how this can be applied investigate tissue-specific age-related activation.
