This review covers a seminal study of the relation between exposure to glucagon-like peptide 1 (GLP-1) agonist semaglutide and incident non-arteritic anterior ischaemic optic neuropathy (NAION) in neuro-ophthalmology clinic setting, subsequent studies unselected populations, meta-analysis clinical trials pathophysiology disc retina that may help elucidate NAION patients with diabetes or obesity. Semaglutide, a glucagon-like peptide-1 receptor agonist, is increasingly used worldwide for its cardiometabolic benefits. However, it has recently been associated with nonarteritic anterior ischemic optic neuropathy (NAION). This report presents four clinical cases and explores possible association. Four male patients were diagnosed NAION during semaglutide treatment, treatment durations of less than one year in three cases. All presented significant disc edema intraretinal fluid on optical coherence tomography, along crowded small Bruch's membrane opening diameters (< 1.4 mm cases). One patient exhibited drusen. Visual field defects corresponded ganglion cell layer atrophy tomography. Systemic risk factors varied; two had only obesity. Our observation suggests that individuals diameter may be at developing treatment.
