Abstract
Silkworm
silk
gland
cells
undergo
multiple
rounds
of
endoreplication,
a
process
in
which
the
genome
is
duplicated
without
cell
division,
leading
to
cellular
polyploidization.
This
results
accumulation
genomic
DNA,
serving
as
foundation
for
rapid
proteins
synthesis.
For
first
time,
we
report
previously
uncharacterized
gene,
SGDAcn
,
silkworm
that
clustered
regularly
interspaced
short
palindromic
repeats
(CRISPR)
/
CRISPR‐associated
nuclease
9‐mediated
knockout

increased
size
and
enhanced
production.

Abstract
We
present
the
first
long-read
de
novo
assembly
and
annotation
of
luna
moth
(Actias
luna)
provide
full
characterization
heavy
chain
fibroin
(h-fibroin),
a
long
highly
repetitive
gene
(>20
kb)
essential
in
silk
fiber
production.
There
are
>160,000
described
species
moths
butterflies
(Lepidoptera),
but
only
within
last
5
years
have
we
begun
to
recover
high-quality
annotated
whole
genomes
across
order
that
capture
h-fibroin.
Using
PacBio
HiFi
reads,
produce
reference
genome
for
this
species.
