Red swamp crayfish, Procambarus clarkii, is the most cultured freshwater crayfish species. It attracts significant research attention due to its considerable economic importance. However, limited availability of genome information has impeded further genetic studies and breeding programs. By utilizing Illumina, PacBio, Hi-C sequencing technologies, we present a more comprehensive continuous chromosome-level assembly for P. clarkii than published one. The final size 4.03 Gb, consisting 2,358 scaffolds with N50 42.87 Mb. Notably, 3.68 corresponding 91.42% genome, was anchored 94 chromosomes. comprises 70.64% repetitive sequences, including 5.21% tandem repeats 65.40% transposable elements. Additionally, total 4,456 non-coding RNAs 28,852 protein-coding genes were predicted in 96.26% annotated. This high-quality not only represents improvement provides insights into unique evolution, but also offers valuable developing aquaculture accelerating breeding.

Abstract The Jonah crab, Cancer borealis, is integral to marine ecosystems and supports a rapidly growing commercial fishery in the northwest Atlantic Ocean. This species also has long history as model for neuroscience that expanded our understanding of central pattern generators, neuromodulation, synaptic plasticity, connectivity neural circuits. Here, we present highly contiguous reference genome crab will provide an essential resource advance fisheries, conservation, biomedical research. Using combination PacBio long-read sequencing Omni-C scaffolding, generated final assembly spanning 691 Mb covering 51 chromosome-length scaffolds 106 additional contigs. Benchmarking Universal Single-Copy Ortholog (BUSCO) analysis indicated high-quality with completeness score 90.8%. Repeat annotation identified 1,649 repeat families making up 48.27% genome. Gene predictions annotated 24,830 protein coding genes 92.3% BUSCO score. family evolution revealed expansion gene associated nervous system function, targeted extensive repertoire genes. not only research but serve foundation investigate adaptation stress population structure support sustainable fisheries management during this time changing environmental conditions

Evolutionary adaptation drives organismal adjustments to environmental pressures, exemplified in the diverse morphological and ecological adaptations seen Decapoda crustaceans, particularly brachyuran crabs. Crabs thrive ecosystems, from coral reefs hydrothermal vents terrestrial habitats. Despite their importance, genetic mechanisms underpinning developmental processes, reproductive strategies, nutrient acquisition remain poorly understood.
