Aging is the greatest non-modifiable risk factor for most diseases, including cardiovascular diseases (CVD), which remain leading cause of mortality worldwide. Robust evidence indicates that CVD are a strong determinant reduced brain health and all-cause dementia with advancing age. also closely linked peripheral cerebral vascular dysfunction, common contributors to development progression all types dementia, largely driven by excessive levels oxidative stress (e.g., reactive oxygen species [ROS]). Abstract The role of senescence in disease contexts is complex, however there considerable evidence that depletion senescent cells improves outcomes a variety particularly related to aging, cognition, and neurodegeneration. Here, the effect bioinformatically-rationalized senolytic was tested experimental autoimmune encephalomyelitis (EAE) mouse model multiple sclerosis (MS). Single-cell analysis from brain tissue isolated mice subjected EAE identified microglia with strong signature including presence BCL2-family member transcripts. Cells expressing Bcl2l1 had higher expression pro-inflammatory genes than their negative counterparts EAE, suggesting they may exacerbate inflammation. Notably, human single-nucleus sequencing MS, BCL2L1 positive were strongly enriched lesions active inflammatory pathology, likewise demonstrated increased immune contribute lesion pathology damage chronic lesions. Employing small molecule BCL2 inhibitor, Navitoclax (ABT-263), significantly reduced spinal cord, these can be targeted by treatment. ABT-263 treatment profound on mice, decreasing motor symptom severity, improving visual acuity, promoting neuronal survival, white matter Together, results provide support idea glia inflammation resulting neuroinflammatory removing them ameliorate disease.

The neutrophil-to-lymphocyte ratio (NLR) is a marker of inflammation that can be obtained quickly, conveniently, and cheaply from blood samples. However, there no research to explore the effects sex age on relationship between NLR mild cognitive impairment (MCI) in community-dwelling older adults.A total 3,126 individuals aged over 60 years Shanghai were recruited for face-to-face interviews, samples collected. MCI was assessed by Mini-Mental State Examination (MMSE) Instrumental Activities Daily Living (IADL) scale, neutrophil count lymphocyte counts measured fasting calculated dividing absolute count. In females, group significantly higher than cognitively normal (2.13 ± 0.94 vs. 1.85 0.83, p < 0.001) but not men. Logistic regression showed an independent risk factor women [odds (OR) = 1.33; 95% confidence interval (CI) 1.14-1.55]. In addition, elevated quartile associated with increased MCI, especially 70 (p value trend 0.012).Compared males, female patients had controls. found years. Therefore, elderly Chinese may target population effective prevention MCI.
