Our skin protects us from external threats including ultraviolet radiation, pathogens and chemicals, prevents excessive trans-epidermal water loss. These varied activities are reliant on a vast array of lipids, many which unique to skin, that support physical, microbiological immunological barriers. The cutaneous physical barrier is dependent specific lipid matrix surrounds terminally-differentiated keratinocytes in the stratum corneum. ABSTRACT Ceramide has transitioned from an incidental discovery to a vital element in skincare, becoming thoroughly studied compound the quest treat skin conditions. Creating moisture barrier, preserving hydration, regulating pH, controlling inflammation, and enhancing functions appearance are among its established benefits. It is often used medically repair barrier defects, as observed inflammatory conditions like atopic dermatitis (AD) dry types. Furthermore, ceramide metabolites commonly predictors before disease manifestation for prognostication processes, thus can be biomarker clinical diagnosis well. In last couple of decades, momentum was also seen pre‐clinical studies involving anti‐cancer nanotechnology field, whereby drug, carrier, or even adjunct formulation increase efficacy treatment such chemotherapy. Approaches levels include directly replenishing lost ceramides with natural extracts, synthetic pseudo‐ceramides, ceramide‐like analogues, well using supplements that stimulate body's production. Although well‐known skincare common AD psoriasis, development related pharmacology severe conditions, cancer, remain stages. Hence, purpose this research explore role health application diseases.

Atopic dermatitis (AD) is a chronic inflammatory skin disease characterized by compromised epidermal barrier and heightened immunoglobulin E (IgE) levels, often associated with filaggrin (FLG) gene mutations. Genetic factors like FLG mutations environmental influences, including microbial exposure pollutants, contribute to the disease’s progression, leading itchy, inflamed skin. AD frequently coexists allergic conditions, severely affecting quality of life. The pathogenesis involves complex interactions between genetic predispositions, immune responses, triggers. Despite advances, development effective treatments remains challenging due an incomplete understanding how influence pathways variability in presentation. Current biomarkers are insufficient fully capture complexity or predict therapeutic highlighting need for novel personalized approaches. Emerging therapies such as chimeric antigen receptor (CAR)-T cell therapy, stem regenerative medicine show promise addressing AD’s root causes. This review explores key aspects pathogenesis, focusing on dysfunction, mechanisms, innovative strategies improve patient outcomes.
