Male hormonal contraception has been the focus of extensive research efforts aimed at identifying effective and reversible methods for male fertility control. This review summarizes current state knowledge, key achievements, future directions in development contraception. A was conducted using PubMed, Embase, Scopus databases. The search strategy included terms such as "male contraception", "Nestorone", "7α,11β-Dimethyl-19-nortestosterone 17β-undecanoate (DMAU)" "11β-methyl-19-nortestosterone 17β-dodecylcarbonate (11β-MNTDC)". total 107 references were analyzed to synthesize most relevant findings regarding contraceptive agents under investigation. outlines historical recent advancements contraception, highlighting compounds that have demonstrated limitations effectiveness, side effects, or inconvenient administration. Notable candidates study include 7α-methyl-19-nortestosterone (MENT), DMAU, 11β-MNTDC, combination segesterone acetate with testosterone gel form. These show promise due their ability suppress follicle-stimulating hormone (FSH) luteinizing (LH), leading spermatogenesis inhibition minimal effects. Additionally, phenomenon spermatogenic rebound is considered. Among investigated agents, oral Nestorone-testosterone appear be promising high efficacy, user-friendly administration, favorable safety profiles. However, further large-scale clinical trials are necessary confirm long-term effects on human health fertility, ensuring viability options.

Abstract Background Dimethandrolone undecanoate (DMAU) is under development as a single agent hormonal male contraceptive. DMAU prodrug hydrolyzed by esterase(s) to the active metabolite dimethandrolone (DMA) which has dual androgenic and progestogenic actions. Phase 1 clinical trial results show be well‐tolerated an oral contraceptive in healthy men; however, delivery of long‐acting injectable rather than daily formulation would provide user compliance benefits address bioavailability concerns. Objective To assess safety, pharmacokinetics (PK), potential non‐human primates (NHP) when delivered or formulation. Materials methods was administered cynomolgus macaques orally for 9 months five weekly intramuscular (IM) injections rhesus IM injection. Evaluations fertility indicators, serum levels DMA were followed > 2 years post‐dose. Results Repeat dose administrations with no significant toxicological findings. Dramatic reductions testosterone concentration occurred within days administration sustained suppression additional indicators (e.g., inhibin B, sperm count, testicular spermatogenesis). Slight body weight increases testes also occurred. resulted concentrations above lower limit quantification (1 ng/mL) 500 days. PK parameters increased increasing dose, while dependence not seen suggesting depot effect reservoir non‐circulating remaining at injection site gets slowly absorbed into circulation.
