Muscle atrophy is a pathological condition characterized by the excessive degradation of muscle proteins, leading to impaired physical performance. Isoliquiritigenin (ISL) promising extract known for its medical effects; however, impact on remains unclear. We investigated effects ISL both in vitro and vivo. The results showed that 5-µM exhibited no significant cytotoxicity C2C12 cells, as reflected cell count kit-8 5-ethynyl-2'-deoxyuridine (EdU) tests. increased diameter myotubes downregulated forkhead box O muscle-specific RING finger protein 1 (MuRF-1), Atrogin-1 induced Dexamethasone (Dex). could also enhance phosphorylation Akt, mammalian target rapamycin (mTOR), eIF4E-binding 1, p70 S6 kinase myotubes. In animal experiments, mass, improved cross-sectional area muscles, inhibited expression MuRF-1 tissues. For performance, enhanced grip strength running endurance. ameliorated Dex-induced vivo, associated with decreased via Akt/mTOR signaling pathway. This suggested be used natural drug atrophy.

Sarcopenia is an ageing-related disease featured by the loss of skeletal muscle quality and function. Advanced glycation end-products (AGEs) are a complex set modified proteins or lipids non-enzymatic glycosylation oxidation. The formation AGEs irreversible, they accumulate in tissues with increasing age. Currently, AGEs, as biomarker ageing, viewed risk factor for sarcopenia. AGE accumulation could cause harmful effects human body such elevated inflammation levels, enhanced oxidative stress, targeted inside outside cells. Several studies have illustrated pathogenic role sarcopenia, which includes promoting atrophy, impairing regeneration, disrupting normal structure extracellular matrix, contributing to neuromuscular junction lesion vascular disorders. This article reviews focused on sarcopenia potential mechanisms detrimental effects, aiming provide new insights into pathogenesis develop novel methods prevention therapy

Abstract Obesity and type-2 diabetes mellitus (T2DM) are interrelated metabolic disorders primarily driven by overnutrition physical inactivity, which oftentimes entails a transition from obesity to T2DM. Compromised musculoskeletal health consistently emerges as common hallmark in the progression of these disorders. Skeletal muscle atrophy dysfunction can further impair whole-body metabolism reduce exercise capacity, thus instigating vicious cycle that deteriorates underlying conditions. However, myocellular repercussions disturbances remain be completely clarified. Insulin signaling not only facilitates skeletal glucose uptake but also plays central role anabolism mainly due suppression catabolic pathways facilitating an anabolic response nutrient feeding. Chronic may trigger different mechanisms proposed contribute insulin resistance aggravate dysfunction. These include inactivation components through sustained activation stress-related pathways, mitochondrial dysfunction, shift glycolytic fibers, hyperglycemia. In present review, we aim delve on mechanisms, providing overview processes involved under chronic overnutrition, their contribution

Background Sarcopenia frequently occurs as a complication among individuals with chronic kidney disease (CKD), contributing to poorer clinical outcomes. This research aimed create and assess predictive model for the risk of sarcopenia in CKD patients, utilizing data obtained from China Health Retirement Longitudinal Study (CHARLS). Methods was diagnosed based on Asian Working Group (AWGS 2019) criteria, including low muscle strength, reduced physical performance, mass. The 2015 CHARLS were split randomly into training set (70%) testing (30%). Forty-nine variables encompassing socio-demographic, behavioral, health status, biochemical factors analyzed. LASSO regression identified most relevant predictors, logistic used explore associated sarcopenia. A nomogram developed prediction. Model accuracy evaluated using calibration curves, while performance assessed through receiver operating characteristic (ROC) decision curve analysis (DCA). Four machine learning algorithms utilized, optimal undergoing hyperparameter optimization evaluate significance factors. Results total 1,092 patients included, 231 (21.2%) Multivariate revealed that age, waist circumference, LDL-C, HDL-C, triglycerides, diastolic blood pressure are significant predictors. These construct nomogram. achieved an AUC 0.886 (95% CI: 0.858–0.912) 0.859 0.811–0.908) validation set. Calibration curves showed good agreement between predicted actual ROC DCA analyses confirmed model’s strong performance. Gradient Boosting Machine (GBM) outperformed other models. Applying Bayesian GBM 0.933 0.913–0.953) 0.932 0.905–0.960) SHAP values age circumference influential Conclusion provides reliable tool predicting patients. exhibits accuracy, positioning it valuable assessment management this population.
