Parkinson's disease (PD) is the second most common age-related neurodegenerative after Alzheimer's disease. Despite numerous studies, specific factors remain unidentified. This study employed a multi-omics approach to investigate link between PD and aging. We integrated blood gene expression profiles, quantitative trait loci, genome-wide association predictive models, conducted clinical validation.By analyzing datasets, total of 953 differentially expressed genes (DEGs) 10 intersecting aging (ADEGs) were identified. Enrichment analysis revealed that regulatory pathways these ADEGs involve classical Wnt signaling pathway, endoplasmic reticulum stress, neuronal apoptosis. Mendelian randomization (MR) showed MAP3K5 significantly reduces risk PD. Multivariate regression identified MXD1, CREB1, SIRT3 as key diagnostic constructed model aid decision-making. Enzyme-linked immunosorbent assay experiments validated levels in serum patients. This utilized identify their mechanisms PD, leading establishment model. An aneurysm is an abnormal enlargement or bulging of the wall a blood vessel. Most often, aneurysms occur in large vessels - aorta (Thoracic Aortic Aneurysm (TAA) and Abdominal (AAA) brain (Intracranial (IA)). Despite presence significant differences pathogenesis development progression IA TAA/AAA, there are also similarities. For instance, both have been shown to be strongly influenced by shear stress, inflammatory processes, enzymatic destruction elastic lamellae extracellular matrix (ECM) proteins vascular wall. Moreover, although TAA predominantly considered arteriopathies with different pathological mechanisms, they share risk factors AAA, such as hypertension smoking. However, need for more in- -depth study key elements that may influence formation particular find ways therapeutic intervention search diagnostic tool. Today, it known disruption gene expression one main mechanisms contribute aneurysms. At same time, growing evidence suggests aberrant epigenetic regulation function related genesis Although much has studied protein-coding genes, circular RNAs (circRNAs), relatively new rapidly evolving family transcripts, recently received scientific attention. CircRNAs regulate through sponging microRNAs (miRNAs) can used targets biomarkers. Cytopathology induced by methamphetamine (METH) is reminiscent of degenerative disorders such as Parkinson’s disease, and it characterized membrane organelles arranged in tubulo-vesicular structures. These areas, appearing clusters vesicles, have never been defined concerning the presence specific organelles. Therefore, present study aimed to identify relative absolute area membrane-bound following a moderate dose (100 µM) METH administered catecholamine-containing PC12 cells. Organelles antigens were detected immunofluorescence, they further quantified plain electron microscopy situ stoichiometry. This analysis indicated an increase autophagosomes damaged mitochondria along with decrease lysosomes healthy mitochondria. Following METH, severe dissipation hallmark proteins from their own vesicles was measured. In fact, amounts LC3 p62 reduced within autophagy vacuoles compared whole cytosol. Similarly, LAMP1 Cathepsin-D reduced. findings suggest loss compartmentalization confirm competence cell clearing during catecholamine degeneration. Such entropy consistent energy stores, which routinely govern appropriate subcellular compartmentalization.
