The influenza A virus (IAV) 2009 H1N1 pandemic was associated with an increased risk of maternal mortality, preterm birth, and stillbirth. underlying mechanism for severe lung disease stillbirth is incompletely understood, but IAV infection known to activate innate immunity triggering the release cytokines. Elucidating impact progesterone (P4), a key hormone elevated in pregnancy, on immune inflammatory response critical step understanding pathogenesis adverse maternal-fetal outcomes. pdm/09 used infect cell lines Calu-3 (lung adenoma) ACH-3P (extravillous trophoblast) or without P4 (100 nM) at multiplicity infections (MOI) 0, 0.5, 3. Cells were harvested 24 48 hours post (hpi) analyzed cytopathic effects (CPE), replicating (TCID50), cytotoxicity (Lactate Dehydrogenase (LDH) assay), NLRP3 inflammasome activation (caspase-1 activity, fluorometric assay). Activation antiviral quantified (RT-qPCR, Luminex) by measuring biomarker gene protein expression ( IFIT1, IFNB ), inflammation IL6 interferon signaling MXA chemokines (IL-8, IL-10). Both highly permissible each timepoint as demonstrated CPE recovery virus. In Calu-3, treatment significant increase cytotoxicity, , IFN-β, IL-6, IL-18. Conversely, ACH-3P, significantly suppressed decreased IFNB, IL1B IFN-β IL-6. both lines, caspase-1 activity after treatment, indicating not higher death Calu-3. summary, these data provide evidence that plays dual role ameliorating viral placenta exacerbating virus-associated injury through nongenomic modulation response.

Abstract Background Animal models of respiratory viral infections are essential for investigating disease pathogenesis and the efficacy antivirals vaccine candidates. A major limitation in research diseases animal is correlating clinically relevant changes pulmonary physiology with cellular molecular mechanistic studies. Few have captured correlated physiologic lung function immune response within same experiment, which critical given heterogeneous nature due to infections. In ventilated human patients, testing can be used not only capture oxygenation, ventilation, but also mechanics yield quantitative measures scalar tracings flow‐volume pressure‐volume loops. Application this protocol during mechanical ventilation non‐human (NHP) would represent a advance research. Methods We applied optimized pigtail macaques ( Macaca nemestrina ) at baseline 5 days after influenza (IAV) inoculation. Results The NHPs manifested clinical hypothermia loss body weight. Declines were striking 66%–81% decline P/F ratio, measure oxygenation reflecting ratio partial pressure oxygen arterial blood (PaO 2 fraction inspiratory concentration (FiO ). There was 16%–45% compliance. Conclusion describe new approach performing primates better correlates demonstrate protection by therapeutics vaccines.

Background. COVID-19 has been a major public health concern globally, leading to higher mortality rate, especially among immunosuppressed individuals, who include pregnant women, people with HIV and living other comorbidities. Pregnant women are considered be special population group owing their specific susceptibility some infectious diseases. Objective. To determine the seroprevalence of anti-SARS-CoV-2 antibodies attending an antenatal clinic. Method. This was descriptive cross-sectional study that tested blood samples from attended clinic March July 2022 using Orient Gene Biotech lateral flow immune-chromatographic assay according manufacturer instructions. The detects IgM IgG against SARS-CoV-2. Results. A total 2 649 were tested; 039 (77.0%) positive for IgG, 7 (0.3%) IgM, 100 (3.7%) both IgM. found 80.7% IgG. Conclusion. findings showed evidence acute infection in our patient despite consensus is dissipating.

ABSTRACT Sexually dimorphic responses to pathogenic infections in animals may stem from sex-specific differences their life history and immune investment. Recent evidence highlights that such variations can also be critically regulated by microbiota. However, direct experiments test how microbiota jointly impacts immunity vulnerability pathogens are still limited. To this end, we used Tenebrio molitor beetles first establish sexes appear differ composition infection responses. Females were more vulnerable bacterial carried a higher load than males. When depleted the microbiome, only females improved post-infection survival, leading loss of outcomes. Males, on other hand, remained unaffected. Microbiota reconstitution (via feeding faecal matter) microbiota-depleted increased susceptibility again, restoring sexual dimorphism. We thus found causal association between microbiome reduced expression an antimicrobial peptide tenecin 1 females, which could associated with susceptibility, but gene-vs-phenotypic associations not consistent across manipulations. Immune strategies required mediate links response might vary manipulations, warranting future investigations.

The aim this review was analyze literature data on effect acute regulation women during pregnancy menopause. We reviewed full-text publications public domain from PubMed, eLibrary.ru, Google Scholar databases with coverage period more than twenty years (2000–2023). Disruptions progesterone estrogen production infection influenza A (H1N1) B viruses have been shown experimental studies. Hormonal status pregnant detected virus (H3N2) depend antiviral antibody titers presence fetoplacental insufficiency. Pregnant COVID-19 may experience brain structure activities, as indicated by high frequency those olfactory sensitivity psychoemotional disorders. During menopause, SARS-CoV-2 leads thyroxine, prolactin, estradiol, testosterone, cortisol, 17-hydroxyprogesterone, dehydroepiandrosterone sulfate.
