Respiratory infection by influenza A virus (IAV) is known to cause systemic inflammation, neuroinflammation, and cognitive impairment. We previously found that experimental with IAV affected oligodendrocyte homeostasis, which was associated altered expression of genes involved in myelin maintenance as well the lipidome. In this study, we sought determine if clemastine, an antihistamine promoting properties, could reverse effects on (OL) specific genes, mitigate infection-induced Male female C57BL/6J mice were randomized into groups based clemastine treatment, infection, sex. Treatment vehicle or (10 mg/kg/d) commenced seven days prior inoculation either saline continued throughout experiment. Body weight measured infection. Spatial learning memory assessed Morris water maze. Sickness behavior measuring burrowing response. Immune cell responses determined flow cytometry, RT-qPCR, antigen recall assays ELISA, viral load RT-qPCR. Hippocampal levels neuroinflammatory (Tnf, Cdkn1a) (Plp1, Mag, Ugt8a) Mice infected developed loss, impaired flexibility, reduced behavior, lung immune infiltration, increased circulating anti-viral IgM IgG T epitopes. Infection hippocampal neuroinflammation decreased myelination. Clemastine treatment resulted earlier recovery loss males for both sexes, but neither Tnf Cdkn1a, nor rescued changes genes. However, mitigated neurocognitive
