Sexually propagated scleractinian corals are in high demand for coral reef restoration. However, threatened reef-building corals, many of the molecular mechanisms related to their reproduction remain largely unknown, which forms a major bottleneck large-scale cultivation sexually reproducing corals. In this study, we analyzed proteomic signatures red eggs and spermatozoa from ecologically significant Galaxea fascicularis, using data-independent acquisition mass spectrometry (DIA-MS) method. A total 7741 7279 proteins mature were identified, respectively. Among these proteins, 596 spermatozoa-specific 1056 egg-specific. Additionally, 4413 differentially abundant (DAPs) among 3121 up-regulated 1292 spermatozoa. Furthermore, anenrichment analyses showed that DAPs identified mainly involved progesterone-mediated oocyte maturation pathway lectin pathway; detected insulin secretion metabolic pathways generation energy. This result will contribute discovery intrinsic regulation gamete fertilization. at least 57 associated with including fluorescent protein (RFP), vitellogenin (VG), egg (EP), testis-specific serine/threonine-protein kinase family (TSSKs), EF-hand Ca2+-binding (EFHC1 EFHC2). Particularly, third yolk EUPHY was reported first time G. fascicularis. conclusion, study unveiled groundbreaking insights into sexual reproduction, paving way more effective conservation sustainable development ecosystems
