Understanding the molecular mechanisms underlying insect resistance to Bacillus thuringiensis (Bt) pesticidal proteins is crucial for sustainable pest management. Here, we found that downregulation of Plutella xylostella ecdysone oxidase gene (PxEO) in normal feeding stages contributes increased 20-hydroxyecdysone (20E) titer and mediates Bt Cry1Ac toxin. The PxEO was cloned its expression significantly downregulated midgut Bt-resistant Cry1Ac-selected P. xylostella. Silencing reduced susceptibility, closely linked protein metabolized (E) 20E vitro, reduction elevated titers activated MAPK-mediated trans-regulatory mechanism known directly cause phenotype. Together with our recently reported 20E-degrading glucose dehydrogenase, this finding highlights a robust, multipronged, approach developed by 20E-mediated defense against harmful agents.

Glyphosate is a widely used non-selective, broad-spectrum, systemic herbicide by interfering with the biosynthesis of aromatic amino acids. However, emergence glyphosate-resistant weeds has driven need for enhanced resistance in crops. In this study, we engineered two mutant variants tobacco EPSPS gene through acid substitution (TIPS-NtEPSPS and P180S-NtEPSPS). These mutated genes were overexpressed under control CaMV35S promoters. Nanoparticles (NPs) exhibit the potential to enhance plant tolerance organic pollutant stress, but how they drive endogenous molecules detoxify contaminants remains be further investigated. This study clarified modulatory mechanisms by which foliar or root application of biosynthesized titanium oxide NPs (g-nTiO2) alleviated atrazine (ATZ) toxicity Lactuca sativa L. Compared with ATZ-alone group, 10 mg/L g-nTiO2 intensified light-harvesting, photoelectron transfer, and reduced oxidative damage, thereby improving growth inducing metabolic reprogramming.
