Abstract Obesity is a complex, chronic disease and global public health challenge. Characterized by excessive fat accumulation in the body, obesity sharply increases risk of several diseases, such as type 2 diabetes, cardiovascular disease, nonalcoholic fatty liver linked to lower life expectancy. Although lifestyle intervention (diet exercise) has remarkable effects on weight management, achieving long-term success at loss extremely challenging, prevalence continues rise worldwide. Over past decades, pathophysiology been extensively investigated, an increasing number signal transduction pathways have implicated obesity, making it possible fight more effective precise way. In this review, we summarize recent advances pathogenesis from both experimental clinical studies, focusing signaling their roles regulation food intake, glucose homeostasis, adipogenesis, thermogenesis, inflammation. We also discuss current anti-obesity drugs, well compounds trials, that target these signals. The evolving knowledge may shed light future direction research, move into new era precision medicine.

Abstract Alpha-melanocyte stimulating hormone (α-MSH) and its receptor, melanocortin 1 receptor (MC1R), have been proposed as potential target for anti-cancer strategies in melanoma research, due to their tissue specific expression involvement melanocyte homeostasis. However, role prevention treatment of is still debated controversial. Although a large body evidence supports α-MSH preventing development, some preclinical findings suggest that the downstream signalling may promote immune escape cancer resistance therapy. Abstract Mammalian hibernators survive prolonged periods of cold and resource scarcity by temporarily modulating normal physiological functions, but the mechanisms underlying these adaptations are poorly understood. The hibernation cycle thirteen-lined ground squirrels (Ictidomys tridecemlineatus) lasts for 5–7 months comprises weeks hypometabolic, hypothermic torpor interspersed with 24–48-h an active-like interbout arousal (IBA) state. We show that squirrels, who endure entire season without food, have negligible hunger during IBAs. These exhibit reversible inhibition hypothalamic feeding center, such arcuate nucleus neurons reduced sensitivity to orexigenic anorexigenic effects ghrelin leptin, respectively.
