ABSTRACT Thiosulfate–citrate–bile salts–sucrose (TCBS) agar is a selective and differential media for the enrichment of pathogenic Vibrios . We observed that an exonuclease VII ( exoVII ) mutant Vibrio cholerae failed to grow on TCBS agar, suggesting DNA repair strains may be hampered growth in this media. Examination components revealed bile acids were primarily responsible toxicity mutant. Suppressor mutations gyrase restored mutants TCBS, inhibits similar antibiotic ciprofloxacin. To better understand what factors are important V. we generated randomly barcoded TnSeq (RB-TnSeq) library have used it uncover range also fail agar. The results study suggest causes damage similarly mechanism action fluoroquinolones, overcoming critical medium. IMPORTANCE often diagnose cholera infection. found many attenuated meaning they could remain undetected using culture-dependent method. Hypermutator with defects pathways might especially inhibited by TCBS. In addition, grown successively develops resistance
