Abstract Although not essential for their growth, the production of secondary metabolites increases fitness producing microorganisms in natural habitat by enhancing establishment, competition, and nutrient acquisition. The Gram-positive soil-dwelling bacterium, Bacillus subtilis, produces a variety metabolites. Here, we investigated regulatory relationship between non-ribosomal peptide surfactin sactipeptide bacteriocin subtilosin A. We discovered that B. subtilis mutants lacking exhibited higher A compared to parental wild-type strain. Additionally, spatial visualization demonstrated secreted colony could suppress an adjacent mutant production. Reporter assays were performed using specific transcriptional regulators, which confirmed role ResD as activator encoding biosynthetic gene cluster (BGC), while removal Rok AbrB repressors increased expression BGC, was further enhanced additional deletion surfactin, suggesting so-far-unidentified regulator might mediate influence on Our study reveals one metabolite another, highlighting function be more complex than its other organisms interactions among also contribute ecological significance.

ABSTRACT Encoded within many microbial genomes, biosynthetic gene clusters (BGCs) underlie the synthesis of various secondary metabolites that often mediate ecologically important functions. Several studies and bioinformatics methods developed over past decade have advanced our understanding both pangenomes BGC evolution. In this minireview, we first highlight challenges in broad evolutionary analysis BGCs, including delineation boundaries clustering BGCs across genomes. We further summarize key findings from comparative genomics on conservation taxa habitats discuss potential fitness effects different settings. Afterward, recent research showing importance genomic context production evolution is highlighted. These draw parallels to recent, broader, investigations gene-to-gene associations pangenomes. Finally, describe mechanisms by which evolve, ranging acquisition or origination entire micro-evolutionary trends individual genes. An outlook how expansions capabilities some might support theories open are result adaptive also discussed. conclude with remarks about future work leveraging longitudinal metagenomics diverse ecosystems likely significantly improve genomes BGCs.

In silico genome mining provides easy access to secondary metabolite biosynthetic gene clusters (BGCs) encoding the biosynthesis of many bioactive compounds, which are basis for important drugs used in human medicine. However, association between BGCs and other functions encoded genomes producers have remained elusive. Here, we present a systems biology workflow that integrates with detailed pangenome analysis detecting genes associated particular BGC. We analyzed 3,889 enterobacterial found 13,266 BGCs, represented by 252 distinct BGC families 347 additional singletons. A revealed 88 putatively specific coding colon cancer-related colibactin code diverse metabolic regulatory functions. The presented opens up possibility discover novel metabolites, better understand their physiological roles, guide identify analyze sets.
