ABSTRACT Objective Migraines profoundly impact patients' quality of life. This study seeks to investigate the relationship between dysregulated miR‐22‐3p and neuroinflammation central sensitization associated with migraine. Methods Initially, level in migraine patients were analyzed using RT‐qPCR. Subsequently, a model was established by administering nitroglycerin (NTG) mice. To modulate levels within this model, agomir utilized. Following intervention, mechanical thermal pain sensitivity evaluated Von Frey filament radiant heat. The c‐Fos, CGRP, TNF‐α, IL‐1β, IL‐6 trigeminal nucleus caudalis (TNC) detected RT‐qPCR ELISA. Furthermore, dual luciferase reporting assays conducted ascertain whether could target KLF6. Moreover, influence KLF6 on inflammatory cytokines further studied. Results significantly reduced NTG In animals, overexpression alleviated hyperalgesia induced NTG. miR‐22‐3p, we observed an increase withdrawal latency, paw threshold, periorbital threshold. Conversely, exhibited significant reduction. We found that can inhibit expression. Additionally, findings indicated suppression resulted decreased along diminished expression IL‐6. Conclusion context migraine, may play pivotal role mitigating alleviating through inhibition
