Genetic studies have revealed hundreds of loci associated with bone-related phenotypes, including bone mineral density (BMD) and fracture risk. However, translating discovered into effective new therapies remains challenging. We review success stories PCSK9-related drugs in cardiovascular disease evidence supporting the use human genetics to guide drug discovery, while highlighting advances artificial intelligence machine learning potential improve target discovery skeletal biology. These strategies are poised how we integrate diverse data types, from genetic electronic health records single-cell profiles knowledge graphs. Such emerging computational methods can position genomics for a future more precise, treatments, ultimately improving outcomes patients common rare disorders.
