In the hippocampus GABAergic local circuit inhibitory interneurons represent only ~10–15% of total neuronal population; however, their remarkable anatomical and physiological diversity allows them to regulate virtually all aspects cellular function. Here we provide an overview current state field interneuron research, focusing largely on hippocampus. We discuss recent advances related various cell types, including development maturation, expression subtype-specific voltage- ligand-gated channels, roles in network oscillations. also technological approaches that have permitted high-resolution, examination numerous neural disorders emerging therapeutic strategies ameliorate such pathophysiological conditions. The ultimate goal this review is not a touchstone for field, but help pave way future research by highlighting where gaps our knowledge exist how complete appreciation will aid strategies.

Many physiologic effects of l-glutamate, the major excitatory neurotransmitter in mammalian central nervous system, are mediated via signaling by ionotropic glutamate receptors (iGluRs). These ligand-gated ion channels critical to brain function and centrally implicated numerous psychiatric neurologic disorders. There different classes iGluRs with a variety receptor subtypes each class that play distinct roles neuronal functions. The diversity iGluR subtypes, their unique functional properties roles, has motivated large number studies. Our understanding advanced considerably since first subunit gene was cloned 1989, research focus expanded encompass facets biology have been recently discovered exploit experimental paradigms made possible technological advances. Here, we review insights from more than 3 decades studies an emphasis on progress occurred past decade. We cover structure, function, pharmacology, neurophysiology, therapeutic implications for all assembled subunits encoded 18 genes. SIGNIFICANCE STATEMENT: Glutamate important virtually aspects either involved mediating some clinical features neurological disease or represent target treatment. Therefore, pharmacology this will advance our many at molecular, cellular, system levels provide new opportunities treat patients.

Myalgic encephalomyelitis/chronic fatigue syndrome (ME/CFS) is a debilitating chronic disease of unknown aetiology that recognized by the World Health Organization (WHO) and United States Center for Disease Control Prevention (US CDC) as disorder brain. The predominantly affects adults, with peak age onset between 20 45 years female to male ratio 3:1. Although clinical features have been well established within diagnostic criteria, diagnosis ME/CFS still exclusion, meaning other medical conditions must be ruled out. pathophysiological mechanisms are unclear but neuro-immuno-endocrinological pattern CFS patients gleaned from various studies indicates these three pillars may key point understand complexity disease. At moment, there no specific pharmacological therapies treat disease, several studies' aims therapeutic approaches described in order benefit patients' prognosis, symptomatology relief, recovery pre-existing function. This review presents approach understanding essential concepts ME/CFS, an emphasis on population, clinical, genetic associated ME/CFS.
