Abstract Despite the lungs being essential for ventilation and aerobic exercise capacity, conventionally are not thought to adapt training. Endurance is key pulmonary rehabilitation programs, which also displays sex-specific differences in therapeutic efficacy. Given molecular underpinnings of lung adaptations endurance uncharacterized, we used a multi-omics approach study sex 6-month-old Fischer 344 rats response an 8 week progressive treadmill training protocol. This was accomplished by reannotating publicly accessible data from Molecular Transducers Physical Activity Consortium (MoTrPAC) integrating newly-analyzed acetylome assess multi-omic sedentary progressively trained states. Female displayed enrichment immune-related features pathways at transcriptome proteome level that were maintained with Conversely, male rat there overall decrease immune following weeks Sexually conserved responses included increased transcriptomic related type I alveoli proteomic cilia, decreased mitochondrial protein acetylation. The impact of cigarette smoke (CS) on lung diseases and the role microbiome dysbiosis in chronic obstructive pulmonary disease (COPD) have been previously reported; however, relationships remain unclear. Our research examined effects 20-week exposure intestinal microbiomes C57BL/6JNarl mice, alongside a comparison with COPD patients' data from public dataset. study found that CS significantly decreased forced vital capacity (FVC), thickened airway walls, induced emphysema. Increased damage was observed along higher keratinocyte chemoattractant (KC) levels by exposure. Lung analysis revealed rise Actinobacteriota, while showed significant diversity changes, indicating dysbiosis. Principal coordinate highlighted distinct compositions between control CS-exposed groups. In microbiome, notable decreases Patescibacteria, Campilobacterota, Defferibacterota, Desulfobacterota were observed. We also identified correlations function both microbiomes. interleukins, interferon-ɣ, KC, 8-isoprostane linked to Notably, patterns mice similar those patients, particularly Global Initiative for Chronic Obstructive Disease (GOLD) stage 4 patients. This suggests systemic summary, induces microbiomes, correlating decline injury. These results align changes underscoring important smoke-related diseases.

Purpose: Chronic obstructive pulmonary disease (COPD) is the main cause of mortality world widely. Non-coding RNAs (lncRNAs) and associated competitive endogenous (ceRNAs) networks were recently proved to lead mRNA gene expression downregulation but still unclear in COPD. This study aims investigate elucidate mechanisms underlying involvement ceRNA co-expression COPD pathogenesis. Methods: Obtained signature data from Gene Expression Omnibus database compared differentially mRNAs miRNAs between patients healthy smokers. Predicted miRNA–lncRNA miRNA–mRNA interaction using online library employed CIBERSORT measure proportions 22 immune cells control groups. Results: Established a ceRNA-network comprising 11 lncRNAs, 5 miRNAs, 16 mRNAs. Abstract Segmental instillation of lipopolysaccharide (LPS) by bronchoscopy safely induces transient airway inflammation in human lungs. This model enables investigation pulmonary inflammatory mechanisms as well pharmacodynamic analysis investigational drugs. The aim this work was to describe the transcriptomic profile segmental LPS challenge with contextualization major respiratory diseases. Pre-challenge bronchoalveolar lavage (BAL) fluid and biopsies were sampled from 28 smoking, healthy participants, followed saline control. Twenty-four hours post instillation, BAL collected challenged lung segments. Total RNA cells biopsy samples sequenced analysed for differentially expressed genes (DEGs). After compared saline, 6316 DEGs upregulated 241 downregulated BAL, but only one DEG samples.
