Abstract Developing nanomedicines with superior reactive oxygen species (ROS) scavenging capability has emerged as a promising strategy in treating ROS‐related diseases, for example, drug‐induced liver injury. However, designing nanoscavengers the self‐propelling ability to scavenge ROS actively remains challenging. Here, self‐propelled silica‐supported ultrasmall gold nanoparticles‐tannic acid hybrid nanozyme (SAuPTB) is designed that can effectively alleviate acetaminophen (APAP)‐induced injury by excessive and regulating inflammation. SAuPTB exhibits multienzyme activity displays significantly enhanced diffusion under hydrogen peroxide (H 2 O ). This vitro research shows eliminate ROS, increasing viability of H ‐stimulated cells reducing cytotoxicity APAP/H ‐treated AML12 cells. The vivo studies show accumulate at inflammatory sites mouse liver, resulting decrease alanine aminotransferase, aspartate reduction pro‐inflammatory cytokines chemokines, hence reduced hepatocyte necrosis, injury, mortality. Furthermore, activates nuclear erythroid 2‐related factor pathway upregulate antioxidative genes reduce oxidative stress. Finally, decreased high mobility group box 1 F4/80 + macrophages, suggesting an anti‐inflammatory response. work provides novel design nanozymes disease treatment.
