Long-term high fructose intake drives oxidative stress, causing glomerular podocyte injury. Polydatin, isolated from Chinese herbal medicine Polygonum cuspidatum, is used as an antioxidant agent that protects kidney function. However, it remains unclear how polydatin prevents stress-driven damage. In this study, attenuated fructose-induced expression of HIF-1α, inhibited NOX4-mediated stromal cell-derived factor-1α/C-X-C chemokine receptor type 4 (SDF-1α/CXCR4) axis activation, reduced reactive oxygen species (ROS) production in rat glomeruli and cultured podocytes. As a result, up-regulated nephrin podocin, down-regulated transient potential cation channel 6 (TRPC6) these animal cell models. Moreover, resveratrol and/or polydatin­derived metabolites solely detected group, indicating these microbial transformed might partly contribute beneficial effects can employed agent improve non-alcoholic fatty disease via metabolism.

The activation of Yes-associated protein (YAP) pathway is mutually causal with the increase extracellular matrix (ECM) stiffness. Polydatin (PD) has been proved to have anti-fibrosis effect in diabetic kidney disease (DKD), but it still a mystery whether PD participates YAP-related mechano-transduction. Therefore, this study intends solve following two problems: 1) To construct an vitro system polyacrylamide hydrogels (PA gels) based on true stiffness kidneys healthy and DKD rats, observe pathological stiffness-induced YAP expression renal fibroblasts; 2) Compared verteporfin (VP), pharmacological inhibitor YAP, explore therapeutic vivo model related regulation YAP. In study, PA gels 3 kPa, 12 kPa 30 was constructed determined for first time simulate rats 8 weeks 16 weeks, respectively. stiffness, significantly increased α-smooth muscle actin (α-SMA) collagen I, production reactive oxygen species (ROS) fibroblasts, were highest. inhibited above-mentioned changes fibroblasts induced by suggesting that inhibition fibroblast-to-myofibroblast transformation ECM at least partially associated regulating mechano-transduction pathway. Importantly, inhibitory nuclear translocation similar VP, superior VP reducing urinary protein, blood glucose, urea nitrogen serum creatinine, as well decreasing α-SMA ROS overproduction fibrosis.
