This study aimed to investigate the effects of silencing Ras homolog family member C (RhoC) on proliferation, apoptosis, invasion, migration, and epithelial-mesenchymal transition (EMT) salivary adenoid cystic carcinoma (SACC) its molecular mechanisms. A total 27 SACC lesions normal gland tissues that were surgically resected at Qingdao Municipal Hospital from January 1, 2019 March 2024 selected, expression levels RhoC detected by Western blot immunohistochemistry. Three small interfering RNA (siRNAs) designed target gene sequence, transfected into SACC-LM SACC-83 cell lines, evaluated for transfection efficiency. The protein RhoC, Rho-associated kinase-1 (ROCK1), p38 mitogen-activated kinase (p38MAPK), phosphorylated-p38MAPK (p-p38MAPK), twist bHLH transcription factor 1 (TWIST1), E-cadherin, N-cadherin, Vimentin compared using blot. CCK-8 assay, flow cytometry, transwell invasion wound healing assay conducted assess differences in migration abilities among groups. Bioinformatics methods also used predict possible upstream micro RNAs (miRNAs) their SACC. Moreover, dual-luciferase reporter experiments performed verify binding sites miR-138-5p RhoC. was highly expressed (P<0.05). After test group showed a significant decrease level ROCK1, p-p38MAPK, TWIST1, Vimentin, as well increase E-cadherin No difference p38MAPK observed (P>0.05). ability decreased group, whereas apoptosis rates significantly increased lowly SACC, mimic can downregulated luciferase activity 293T cells after with wild-type plasmid is may downstream ROCK1/p38MAPK/TWIST1 signaling pathway, thereby inhibiting EMT while promoting apoptosis. On contrary, potential there be between two genes.

Background/Objectives: Salivary gland carcinomas encompass a broad group of malignant lesions characterized by varied prognoses. Stem cells have been associated with the potential for self-renewal and differentiation to various subpopulations, resulting in histopathological variability diverse biological behavior, features that characterize salivary carcinomas. This study aims provide thorough systematic review immunohistochemical studies regarding expression prognostic significance stem cell markers between different tumors (MSGTs). Methods: The English literature was searched via databases MEDLINE/PubMed, EMBASE OVID, Web Science, Scopus, CINHAL EBSCO. Joanna Briggs Institute Critical Appraisal Tool used risk bias (RoB) assessment. Meta-analysis conducted evaluated same pair diseases at least two studies. Results: Fifty-four reported markers, e.g., c-KIT, CD44, CD133, CD24, ALDH1, BMI1, SOX2, OCT4, NANOG, MSGTs. Low, moderate, high RoB observed twenty-five, eleven, eighteen studies, respectively. revealed an outstanding discriminative ability c-KIT adenoid cystic carcinoma (AdCC) over polymorphous adenocarcinoma [P(LG)A] but did not confirm Conclusions: indicated possible link heterogeneity behavior might be diagnostic value discriminating AdCC P(LG)A.
