ProAKAP4 is a sperm structural protein that regulates motility through the PKA-dependent cAMP signaling pathway, which synthesized as an X chromosome-linked member of gene family. This study aims to determine optimal level proAKAP4 for evaluating sexed semen investigating its relationship with longevity quality in Holstein bull sperm. A total 30 samples (bearing chromosomes) from distinct bulls (n = 30) were analyzed. The frozen assessed their levels, mitochondrial membrane potential, plasma and acrosome integrity (PMAI), spermatozoa movement parameters at hours 0 3 after thawing. levels ranged 16.35 72.10 ng/10 M spz, average 37.18 ± 15.1 spz. strong positive correlation was observed between motility, progressive PMAI, high VAP, VCL values h incubation, when compared post-thaw analyses. results also reveal ≥40 spz exhibit higher quality.
