Background Atherosclerosis (AS) is a chronic inflammatory disease characterized by endothelial dysfunction, monocyte infiltration, smooth muscle proliferation, and extracellular matrix accumulation. Endothelial cell (EC) dysfunction plays pivotal role in the initiation progression of AS. Despite progress traditional research methods, complexity cellular heterogeneity within remains poorly understood, necessitating more refined approach for uncovering mechanisms. Methods In this study, we employed single-cell RNA sequencing (scRNA-seq) to map landscape AS comprehensively. By analyzing heterogeneity, differentiation trajectories, functional states, identified critical subpopulations their roles Functional enrichment analyses were conducted, findings validated through vitro experiments. Results The analysis revealed distinct EC with unique contributions progression. Among these, C1 CXCL12 + ECs emerged as key subpopulation associated differentiation, vascular remodeling, inflammation. These cells demonstrated high proliferative potential enriched pathways related migration repair. Through CCK-8, Transwell assay, EdU staining angiogenesis ability, found that knockdown FOXM1 CXCL12+ resulted decreased invasion. Thus, it affects Conclusion This study provides detailed atlas AS, identifying subpopulations, regulatory pathways, factors driving application technologies paves way advancing our understanding cardiovascular diseases offers significant developing personalized therapeutic strategies immunology precision medicine.

Abstract Colorectal cancer (CRC) is a relatively common malignancy clinically and the second leading cause of cancer‐related deaths. Recent studies have identified T‐cell exhaustion as playing crucial role in pathogenesis CRC. A long‐standing challenge clinical management CRC to understand how T cells function during its progression metastasis, whether potential therapeutic targets for treatment can be predicted through cells. Here, we propose DeepTEX, multi‐omics deep learning approach that integrates cross‐model data investigate heterogeneity DeepTEX uses domain adaptation model align distributions from two different modalities applies cross‐modal knowledge distillation predict across diverse patients, identifying key functional pathways genes. offers valuable insights into application multi‐omics, providing exploring stages associated with relevant targets.

Prostate cancer (PCa) is a multifactorial and heterogeneous disease, ranking among the most prevalent malignancies in men. In 2020, there were 1,414,259 new cases of PCa worldwide, accounting for 7.3% all malignant tumors. The incidence rate ranks third, following breast lung cancer. Patients diagnosed with high-grade frequently present existing or developing metastases, complicating their treatment resulting poorer prognoses, particularly those bone metastases. Utilizing single-cell RNA sequencing (scRNA-seq), we identified specific cell subtypes that are closely linked to PCa. By investigating mechanisms govern interactions within tumor microenvironment (TME), aim offer theoretical insights can enhance prevention, diagnosis, PCa, ultimately striving improve patient outcomes quality life.
