Intermittent fasting (IF) is known to induce significant ischemic tolerance. Diet a major proponent of gut microbiota, and microbial dysbiosis plays role in post-stroke brain damage. Hence, we currently evaluated whether IF-mediated tolerance mediated by microbiota. Additionally, circadian cycle modulate post-ischemic outcomes, thus further if microbiota would be influenced prophylactic IF during the inactive phase (fasting daytime; IIF) or active nighttime; AIF). The AIF, but not IIF, cohort showed significantly decreased fecal Firmicutes/Bacteroidetes ratio compared with ad libitum (AL) cohort. Moreover, levels microbiota-derived metabolites butyrate propionate AL following focal ischemia, whereas they increased AIF However, transplantation (FMT) from IIF had no effects on motor cognitive function recovery, anxiety-, depression-like behaviors FMT Furthermore, did influence infarct volume, atrophy volume white matter Overall, current findings indicate that beneficial after ischemia are

Cerebral ischemic preconditioning offers a promising strategy to enhance resilience severe insults. Unilateral common carotid artery occlusion (UCCAo) is valuable model simulate chronic cerebral hypoperfusion (CCH). This study explored UCCAo-induced CCH as stimulus induce tolerance against transient global ischemia (tGCI) induced by bilateral (BCCAo) in both male and female mice. We evaluated the effects of on neuroprotection recovery through behavioral, histopathological, molecular analyses. Laser Doppler Imaging (LDI) confirmed significant post-UCCAo. Preconditioning reduced mortality rates at days 1 7 post-surgery compared BCCAo, suggesting its neuroprotective potential. Neurodeficit scoring demonstrated protection preconditioned animals with aligning closer sham controls. Behavioral assays revealed improved motor cognitive outcomes groups, sex-specific differences evident dynamics. Molecular analyses indicated reactive astrocyte (GFAP) microglial (IBA1) activation animals, reflecting controlled glial responses. Sex-dependent variations markers hypoxia (Hif1a), autophagy (Becn1), neurogenesis (Sox2) highlighted neuroadaptive cellular influences resilience. enhanced synaptic plasticity upregulating PSD-95, synaptophysin BDNF levels. Intermittent fasting (IF) has demonstrated extensive health benefits through the regulation of fatty acid metabolism and modulation neuroimmune microenvironment, primarily via activation key signaling pathways such as AMP-activated protein kinase (AMPK) sirtuin 1 (SIRT1). IF not only facilitates oxidation improves metabolic health, but also enhances mitochondrial function, mitigates oxidative stress, promotes autophagy, inhibits apoptosis ferroptosis. These mechanisms contribute to its substantial preventive therapeutic potential in various conditions, including neurodegenerative disorders Alzheimer’s Parkinson’s diseases, autoimmune neurotraumatic conditions. While supportive evidence been obtained from animal models preliminary clinical studies, further large-scale, long-term randomized controlled trials are imperative establish safety evaluate efficacy comprehensively.
