Background Neuroblastoma (NB) primarily arises in children who are <10 years of age, and originates from developing sympathetic nervous system, which results tumors adrenal glands and/or ganglia. The diagnosis NB involves a combination laboratory imaging tests, biopsies. Small extracellular vesicles (sEVs) have gained attention as potential biomarkers for various types tumors. Here, we performed proteomic analysis serum sEVs identified NB. Methods Label-free proteomics were the discovery phase. A bulk RNA-seq dataset tissues was used to analyze association between genes encoding proteins prognosis. Potential validated via multiple reaction monitoring (MRM) or western blot validation public single-cell (scRNA-seq) integrated tissue origin harboring biomarkers. Results total 104 differentially expressed patients with label-free proteomics, 26 MRM analysis. Seven BSG, HSP90AB1, SLC44A1, CHGA, ATP6V0A1, ITGAL SELL showed strong ability distinguish healthy controls non-NB well. Integrated scRNA-seq revealed that these sEVs-derived originated different cell populations tumor tissues. Conclusion sEVs-based may aid molecular NB, representing an innovative strategy improve detection management.

Extracellular vesicles (EVs) are heterogeneous nanoparticles (30–500 nm) found in all biological fluids. The composition of EVs largely reflects the cells that secreted them. At present, widely used to search for diagnostic and therapeutic markers, therefore studies properties becoming more demand today. Despite obvious potential biomedical applications, lack unified methods their isolation, analysis quantitative measurement significantly limits scientific progress this field. In literature there descriptions dozens isolation study EVs, often researchers do not present comparative advantages disadvantages used. regard, a review becomes relevant. This presents comprehensive modern EVs. Each method has its own limitations affecting yield, purity characteristics isolated vesicles, as well accuracy results obtained. choice should be made considering purpose specificity study, since is no universal approach at present.
