Abstract Osteoporosis (OP) is a severe global health issue that has significant implications for productivity and human lifespan. Gut microbiota dysbiosis been demonstrated to be closely associated with OP progression. Melatonin (MLT) an important endogenous hormone modulates bone metabolism, maintains homeostasis, improves Multiple studies indicated MLT participates in the regulation of intestinal gut barrier function. However, promising effects microbiota‐derived remain unclear. Here, we found resulted tryptophan disorder decreased production MLT, while administration could mitigate OP‐related clinical symptoms reverse dysbiosis, including diversity microbiota, relative abundance many probiotics such as Allobaculum Parasutterella , metabolic function flora amino acid nucleotide energy metabolism. Notably, significantly increased short‐chain fatty acids trimethylamine N‐oxide‐related metabolites. Importantly, modulate dynamic balance M1/M2 macrophages, reduce serum levels pro‐inflammatory cytokines, restore gut‐barrier Taken together, our results highlighted roles microbially derived progression via “gut‐bone” axis SCFA which may provide novel insight into development drug treating OP.

Objectives In China, osteoporosis has become a major health concern among elderly population, imposing significant burden on the country’s social and economic systems. The monocyte to high-density lipoprotein ratio (MHR) been currently recommended as novel marker of inflammation oxidative stress associated with in type 2 diabetes mellitus (T2DM). However, its reliability non-diabetic populations remains unclear. present study was evaluate association between MHR population. Methods clinical data 240 subjects (115 group 125 normal bone group) were retrospectively analyzed all statistical analyses performed by using SPSS 26.0. Results Differences age, neutrophils, lymphocytes, monocytes, MHR, uric acid, creatinine, triglycerides, and cholesterol found be statistically two groups. A binary logistic regression model conducted including UA Cr variables. results showed that age an independent risk factor protective for abnormality ROC analysis area under curve predictive effect their combined test 0.623, 0.728 0.761, respectively; correlation positively correlated lumbar hip BMD, negatively femoral neck ratio, intertrochanteric stem showing differences (P<0.05). Conclusions For population: is against abnormalities significantly higher than abnormal group.

Recent studies have confirmed that melatonin and N6-methyladenosine (m6A) modification can influence bone cell differentiation formation. Melatonin also regulate a variety of biological processes through m6A modification. Heterogeneous nuclear ribonucleoprotein A2/B1 (hnRNPA2B1) serves as reader In this study, we used the hindlimb unloading model an animal loss induced by simulated microgravity 2D clinorotation to simulate environment for cells on ground. We found hnRNPA2B1 was downregulated both in vitro vivo during microgravity. Further investigations showed could promote osteoblast overexpression attenuated suppression discovered expression under Moreover, promotion partially dependent hnRNPA2B1. Therefore, research revealed, first time, role melatonin/hnRNPA2B1 axis Targeting may be potential protective strategy against microgravity-induced osteoporosis.
