Trehalose-6-phosphate synthase (HcTPS) and trehalose-6-phosphate phosphatase (HcGOB) are key enzymes for trehalose synthesis in Haemonchus contortus. In addition, previous studies have also demonstrated that HcTPS HcGOB can regulate the function of host immune cells vitro, important immunosuppressive molecules. Therefore, this study evaluated potential as vaccine candidates through vitro vivo experiments. To evaluate inhibitory effects polyclonal antibodies on egg hatching larval development, anti-rHcTPS anti-rHcGOB were incubated separately with eggs first-stage larvae (L1s) under controlled conditions. For immunization studies, recombinant proteins (rHcTPS rHcGOB) formulated Quil-A adjuvant, administered to goats subcutaneous injection. Vaccine efficacy against contortus infection was determined comprehensive analysis multiple parasitological parameters, including: (1) abnormality rate, (2) success (3) reduction output rates, (4) adult worm burden. The results experiments showed had no effect rate eggs, but significantly affected development from L1s infectious third stage (L3s). After protein (rHcTPS) (rHcGOB), high levels antigen-specific immunoglobulin G (IgG) produced goats, remained till end experiment. Compared adjuvant control group, number deformed rHcTPS protein- immunized group rHcGOB increased. protein-immunized deformity 9.59% 17.30%, respectively, reduced by 11.27% 13.71%, respectively. Moreover, compared adults decreased 64.47% 60.93%, 63.97% 69.54%, Furthermore, (Quil-A adjuvant), content reduced. These findings indicate exhibit superior protective effects, rendering them promising development.

Abstract Background: Benzimidazole (BZ) resistance remains an emerging grave concern in helminths of public and veterinary health concerns. Resistance against BZ drugs is due to mutations that change the amino acid comprising β-tubulin protein, which negatively affects its interactions with drug molecules. Several silico modeling studies have been published decipher precise mechanism resistance, but inconsistencies on consequence confer effect different ligands led further confusion regarding exact resistance. Hence, this scoping review was done unravel based research molecular docking dynamics. Methods: A conducted ScienceDirect, MEDLINE via PubMed Scopus using search term “Benzimidazole AND Beta Tubulin Molecular Docking”. Results: The six papers studied several helminth species: Haemonchus conturtos, Trichinella spiralis, Ancylostoma duodenale, caninum, ceylanicum, Necator americanus, Trichuris trichiura, suis, Anisakis simplex, Ascaris suum, Ascaridia galli, Parascaris equorum, Toxocara canis, Fasciola hepatica. benzimidazole resistance-associated F167Y (TTC, TTT → TAC, TAT), E198A (GAG, GAA GCG, GCA), F200Y TAT). results show can markedly reduce binding affinity ligand-β-tubulin interactions. also showed a similar could vary species. mutation alter conformation active site, affecting binding.
