This study aimed to establish a rat model of chronic wounds observe the effects hyperbaric oxygen (HBO) on wound repair and pyroptosis explore potential role in pathogenesis wounds. Sprague-Dawley (SD) rats were randomly divided into acute group (control group), (model + HBO treatment (HBO VX-765 (IL-converting enzyme/Caspase-1 inhibitor) (VX-765 group). After 7 days respective interventions, healing status was observed, tissue specimens collected. Hematoxylin eosin (HE) staining used pathological changes tissues. Transmission electron microscopy cellular ultrastructure. Immunofluorescence expression localization vascular endothelial growth factor A (VEGF-A) N-terminal domain gasdermin D (GSDMD-N). Western blot conducted detect nucleotide-binding oligomerization domain-like receptor protein 3 (NLRP3), cysteine-requiring aspartate protease-1 (Caspase-1), VEGF-A, GSDMD-N proteins Real-time quantitative reverse transcription polymerase chain reaction (qRT-PCR) NLRP3, Caspase-1, GSDMD genes. Enzyme-linked immunosorbent assay (ELISA) performed inflammatory cytokines interleukin-1 beta (IL-1β) IL-18. The results showed that had faster rate better pathology improvement compared group. level VEGF-A higher group, while levels GSDMD, IL-1β, IL-18 lower than those can effectively promote wounds, regulation may be one its mechanisms action. Abstract Despite the efficacy of antiretroviral therapy (ART) in reducing global incidence vertical HIV transmissions, more than 120,000 children are still infected with virus each year. Since ART cannot clear reservoir that is established soon after infection, living (CLWH) forced to rely on for their lives and suffer from long-term drug-related complications. Pediatric like adult associated gut microbial dysbiosis, loss epithelial integrity, bacterial translocation, CD4 + T cell depletion, systemic immune activation, viral establishment. However, unlike adults, vertically acquired by infants interacts a microbiome continuously evolving while concomitantly shaping infant’s ontogeny. Therefore, determine whether there may be interventions target through microbiome-directed approaches, understanding complex tripartite interactions between transmitted HIV, maturing microbiome, developing system during early life crucial.
