Oral submucous fibrosis (OSF) is a chronic oral mucosal disease, which exerts profound impact on patients' daily life and currently lacks efficacious therapeutic interventions. Epigallocatechin-3-gallate (EGCG), the abundant polyphenol found in green tea, exhibits remarkable anti-fibrotic effects skin. However, research OSF regarding EGCG relatively limited. We aimed to investigate potential effect of against using an arecoline (ARE) -induced rat model primary fibroblasts. Primary fibroblasts (ROMF) were isolated identified. Optimal ARE concentrations established Cell Counting Kit-8. The extracellular matrix (ECM)-related protein expression was assessed through RT-qPCR Western blot techniques. Similarly, ARE-induced ECM changes ROMF evaluated. study also Sprague-Dawley rats, induced by ARE, with pathological characterized HE Masson's staining, further assessing ECM-related tissues methods. effectively suppressed components while concurrently improving process vitro vivo. results indicate that natural product increased improved process, indicating could be potentially novel candidate drug for treatment OSF.

Background: The management of OSMF aims to alleviate the signs and symptoms, such as burning sensation release fibrotic bands assist oral opening. Surgical excision is best choice in treatment advanced OSMF. Interpositional grafts are used cover raw surgical defects following bands' excision. They can be local (tongue flap, palatal island buccal fat pad, nasolabial temporalis fascia flap) or distant (abdominal dermal graft, anterolateral thigh flap). This systematic review was conducted evaluate which type interpositional graft effective submucous fibrosis. Material method: A Literature search performed 12 electronic databases for articles published between 2000, August 31, 2023. Two reviewers independently reviewed articles. Ten (Non randomized controlled trial (NRCT)-5 (RCT)-5) were selected based on inclusion criteria. For NRCTs, Joanna Briggs Institute Critical Appraisal Checklist Quasi-Experimental Studies ROB b tool RCT risk bias. Results: studies exclusion It included; 5 NRCTs RCTs. Meta-analysis performed, mouth opening greater subjects treated with flap than other designs at sixth month 12th follow up. However, difference two groups statistically non-significant ( p = 0.19). Buccal Fat pad also shows successful results but not significant. Conclusion: observations this help enhance awareness among healthcare professionals, clinicians about recommending an cases making use its advantages.

Abstract Oral submucous fibrosis (OSF) is a chronic progressive disease associated with increased collagen deposition and TGF-β1 release. The current therapy management have been limited success due to low efficacy adverse drug reactions. This study aimed evaluate epigallocatechin 3-gallate (EGCG) encapsulated nanoparticles loaded mucoadhesive hydrogel nanocomposite (HNC) for OSF. Developed HNC formulations were evaluated their permeation behaviour using in vitro as well ex vivo studies, followed by evaluation of safety studies areca nut extract-induced OSF rats. condition OSF-induced rats was assessed mouth-opening biochemical markers. optimized polymeric exhibited the required particle size (162.93 ± 13.81 nm), positive zeta potential (22.50 2.94 mV) better strength (0.40 0.002 N), faster interactions positively charged surface negatively buccal mucosal membrane. significantly improved conditions reducing concentration without showing toxicity reverting fibroid mucosa normal.
