Geldanamycin, a benzoquinone ansa antibiotic, has been extensively applied in medical, agricultural, and health research areas due to its antitumor, antifungal, herbicidal, antiradiation effects. In this study, an improvement of geldanamycin production by Streptomyces geldanamycininus FIM18-0592 was first performed protoplasts combined with UV mutagenesis ribosome engineering technology, respectively. The results showed that strains induced were superior treated erythromycin terms the positive variability, average relative titer, maximum values 51.95%, 99%, 136%, A mutant strain produced 3742 μg/mL generated protoplast mutagenesis, 36% higher yield than initial strain. Multi-omic analysis revealed high-yielding 53 could upregulate GdmG GdmX 1.59 2.38 times ansamycin synthesis pathway, downregulate pks12, pikAI, pikAII 0.25, 0.37, 0.48 fatty acid which crucial for production. Our study provides novel S. strategy offers valuable insights breeding other microorganisms.
