BACKGROUND The GALAD score has improved early hepatocellular carcinoma (HCC) detection rate. role of the in staging and predicting tumor characteristics or clinical outcome HCC remains particular interest. AIM To determine diagnostic/prognostic performances at various phases initial diagnosis, features, 1-year mortality compare performance with those other serum biomarkers. METHODS This prospective, study was conducted among patients newly diagnosed liver center Vajira Hospital. Eligible had allocation using Barcelona Clinic Liver Cancer (BCLC) categorization. Demographics, etiology, features were recorded. Biomarkers obtained baseline. biomarkers prospectively assessed. RESULTS Exactly 115 individuals HCC. increased disease severity. Between BCLC-0/A BCLC-B/C/D, predicted an area under curve (AUC) 0.868 (95%CI: 0.80–0.93). Abstract Liver hepatocellular carcinoma (LIHC) strongly contributes to global cancer mortality, highlighting the need for a deeper understanding of its molecular mechanisms enhance patient prognosis and treatment approaches. We aimed investigate differential expression immunogenic cell death-related genes (ICDRGs) cellular senescence-related (CSRGs) in LIHC their effects on prognosis. combined GSE25097, GSE46408, GSE121248 datasets by eliminating batch standardizing data. After processing, 16 were identified as ICDR&CSR differentially expressed (ICDR&CSRDEGs), including UBE2T , HJURP PTTG1 CENPA FOXM1 . Gene set enrichment analysis indicated strong these pre-Notch processing. variation revealed 20 pathways with significant differences between control groups. Mutation TP53 most commonly mutated gene samples. A prognostic risk model integrating 12 ICDR&CSRDEGs was developed, showing high precision at 1 year but diminished accuracy 2 3 years. Our constructed provides valuable insights predicting outcomes may guide future therapeutic interventions targeting specific genes. Further research is needed explore mechanistic roles progression response.

Hepatocellular carcinoma (HCC) is one of the deadliest malignant tumors in world, and its incidence mortality have increased year by year. HCC research has increasingly focused on understanding pathogenesis developing treatments. The Wnt signaling pathway, a complex evolutionarily conserved signal transduction system, been extensively studied genesis treatment several tumors. Recent investigations suggest that may be significantly influenced dysregulated Wnt/β-catenin signaling. This article aimed to examine pathway controls mechanisms. In addition, we highlighted role this etiology targeted treatment.

Background: Liver cancer, specifically hepatocellular carcinoma (LIHC), ranks as the second most common cause of cancer-related fatalities globally. Moreover, occurrence rate LIHC is steadily increasing. A recently identified gene, SPSB2, has been implicated in cell signaling, impacting development and progression non-small lung cancer. Nevertheless, studies on role SPSB2 pathogenesis are lacking. Methods: Using TCGA, GTEx, GEO databases, we obtained differentially expressed genes that affect prognosis patients with LIHC. We utilized Kruskal–Wallis test, along univariate multivariate COX regression analyses, to determine correlation between patient clinical indicators. Potential biological functions were explored by enrichment analysis, ssGSEA, Spearman analysis. Finally, lines Huh7 SMMC-7721 used validate function SPSB2. Results: The results showed higher expression had a poorer prognosis, was significantly correlated patients’ Histologic grade, Pathologic T stage, Prothrombin time, BMI, weight, adjacent hepatic tissue inflammation, AFP level, OS event (p < 0.05). shows notable pathways linked tumorigenesis immune system. its strongly connected cells checkpoints. Knockdown inhibits SPSB2’s functions, including proliferation, invasion, metastasis, other phenotypes. Conclusions: plays crucial It related response unfavorable outcomes. may biomarker for prognosis.

ABSTRACT Osteosarcoma (OS) is the most common malignant tumor of bone. This paper aimed to explore mechanism macrophage polarization and glycolysis in OS. Gene expression microarray GSE42572 for OS was downloaded from GEO database validated TCGA‐SARC. CDKN2A cell lines normal human bone osteoblasts detected. Saos2 cells were transfected with siRNA‐CDKN2A, U2OS pcDNA3.4‐CDKN2A knock down or upregulate role behaviors. After co‐incubation macrophages, CD206‐positive decreased, CD86‐positive increased, increased. enriched expression. Upregulation M2 polarization. downregulation growth, metabolic reprogramming, enhances its expression, which turn activates polarization, ultimately promoting progression.
